I was having tremendous difficulty in eliminating the "unlicensed" message that was appearing, despite following the instructions. After a few hours of trial and error, I discovered that using the wildcard assembly version in the AssemblyInfo file, e.g. [assembly: AssemblyVersion("5.0.*")] seemed to be the cause. Is this correct? And if it is correct, then each time we create an updated version do we have to a) manually increment the version number, and b) manually generate and update the license key?

The version number should be generated automatically. About the license key, yes, you need to re-generate the runtime license key everytime you make a major change.
